Several techniques are used for quantification. Enzymatic recycling assays rely on glutathione reductase and a colorimetric or fluorescent readout, offering sensitivity for total glutathione. High-performance liquid chromatography can separate GSH from GSSG and other thiols, often with UV, fluorescence, or electrochemical detection. Mass spectrometry provides structural confirmation and can quantify low-abundance species when paired with separation. Each approach has trade-offs in specificity, throughput, and equipment requirements, so method selection depends on the research question and available instrumentation.
Stability depends on pH, temperature, oxygen exposure, and trace metals. Aqueous solutions of reduced glutathione are susceptible to oxidation, especially when neutral or alkaline and exposed to air. Transition metal ions can catalyze thiol oxidation, so chelators and inert atmospheres are sometimes used in research settings. Standards are typically stored cold and desiccated, with limited freeze-thaw cycles. Questions remain about how closely in vitro stability data reflect the behavior of glutathione within intact cells and tissues.
Glutathione functions in redox balance, detoxification, and sulfur amino acid storage. It participates in reactions that help maintain ascorbate and protein thiol status. The molecule serves as a cofactor for several enzymes, including glutathione peroxidases and glutathione S-transferases. These enzymes reduce peroxides and conjugate electrophiles, respectively. Glutathione also contributes to the metabolism of xenobiotics and to the transport of cysteine between tissues. How interorgan transport and tissue-specific regulation shape whole-body pools remains an active area of study.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. Its cysteine residue carries a thiol group, which allows the molecule to participate in reduction and oxidation reactions. The compound exists in most living cells, where the reduced form, often abbreviated GSH, is usually more abundant than the oxidized disulfide form, GSSG. Intracellular concentrations are commonly in the millimolar range, while extracellular concentrations are much lower. This uneven distribution supports its role as a major cellular redox buffer.
Cells synthesize glutathione through two ATP-dependent enzymatic steps. The first step combines glutamate and cysteine to form gamma-glutamylcysteine, catalyzed by glutamate-cysteine ligase. The second step adds glycine, producing the complete tripeptide, catalyzed by glutathione synthetase. Glutathione itself can inhibit the first enzyme, providing negative feedback when levels are high. Because cysteine is often limiting, its availability influences how quickly the pathway proceeds. These reactions occur in the cytosol, and the resulting glutathione can be distributed to other compartments.

| Reduced form | GSH | Main intracellular thiol |
| Oxidized form | GSSG | Disulfide dimer of two GSH molecules |
| Common separation method | Reversed-phase HPLC | Often with ion-pairing or derivatization |
| Typical detection | Fluorescence or mass spectrometry | UV detection is also used in some assays |
| Storage of standards | -20 °C or below, desiccated | Limit freeze-thaw and moisture exposure |
Storage recommendations for glutathione reagents usually specify a cool, dry, dark environment because the thiol oxidizes in air and light. Solid material is often kept desiccated at low temperature, while solutions are prepared fresh or stored frozen in aliquots. Repeated freeze-thaw cycles can accelerate degradation, and metal ions can catalyze oxidation. Quality control may include purity assays, water content, and identity confirmation. Stability limits are method-specific, so a stated shelf life applies only to defined conditions and packaging.
Laboratory measurement of glutathione requires attention to oxidation before analysis. Blood, tissue, or cell samples can lose reduced glutathione as it converts to GSSG or forms mixed disulfides with proteins. Acid extraction, rapid freezing, and thiol-blocking reagents are common strategies to preserve the original distribution. Reported concentrations therefore depend on collection protocol, extraction method, and the time between sampling and analysis. Comparisons across studies are most reliable when these pre-analytical variables are described.
Biosynthesis proceeds in two ATP-dependent steps. First, glutamate-cysteine ligase joins glutamate and cysteine. Second, glutathione synthetase adds glycine to the intermediate. The pathway is regulated by cysteine availability, enzyme expression, and feedback inhibition by glutathione itself. Liver tissue has a particularly high capacity for synthesis and export. Because the molecule is made inside cells, circulating glutathione reflects a balance of release, uptake, and breakdown rather than simple dietary supply.
Functionally, glutathione supports redox balance by donating electrons and becoming oxidized. It also serves as a cofactor for enzymes such as glutathione peroxidases and glutathione S-transferases. These enzymes participate in peroxide reduction and in conjugation reactions that help process reactive molecules. Separate from antioxidant roles, glutathione can modify protein cysteines through S-glutathionylation, influencing enzyme activity and signaling. Research continues to examine how these chemical roles translate into whole-organism effects.
Glutathione is a tripeptide composed of glutamate, cysteine, and glycine. Its glutamate-cysteine linkage uses the gamma-carboxyl group of glutamate, a feature that resists standard peptidases. The cysteine residue provides a thiol group, which gives the molecule its reducing character. In cells, glutathione is often the most abundant small-molecule thiol, with concentrations varying widely by tissue and compartment. It exists mainly in a reduced form called GSH, while oxidation produces a disulfide-linked dimer called GSSG.
"... the death penalty is primarily a criminal justice issue, and therefore is a question for the sovereign jurisdiction of each country [...] the right to life is not the only right, and [...] it is the duty of societies and governments to decide how to balance competing rights against each other." In January 2004, the Ministry of Home Affairs issued a response to Amnesty International's report, "Singapore: The death penalty - A hidden toll of executions". It defended Singapore's policy to retain the death penalty, predicating its arguments on, among others, the following grounds:
A medical specialty is a branch of medical practice that is focused on a defined group of patients, diseases, skills, or philosophy. Examples include those branches of medicine that deal exclusively with children (pediatrics), cancer (oncology), laboratory medicine (e.g. pathology), or primary care (family medicine). After completing medical school or another form of basic training, physicians or surgeons and other clinicians usually further their medical education in a specific specialty of medicine by completing a multiple-year residency to become a specialist.
And while it had been speculated that the availability of such treatment options might change public perception of the risks associated with drug use and might lead to an increase in illicit drug use, the incidence of heroin abuse in Switzerland has declined sharply since the introduction of heroin-assisted treatment. As a study published in The Lancet concluded:
=== Intellectual property === The first polio vaccine was never patented; some have argued that similar treatment of an effective COVID‑19 vaccine could enable fair distribution. Initially, negotiations at the World Trade Organization (WTO) on the issue of waiving patent rights were blocked for months by resistance by the US, Switzerland, Norway, and the EU. Initially one observer considered the US position unlikely to change, but as of April 2021 the US administration was discussing the issue and then reversed course and announced its support for a patent waiver for COVID-19 vaccines on 5 May 2021. 400 non-profit organizations and 115 members of the European Commission have signed a letter urging the United States and Europe to side with the WTO members in the global south.
=== Reasons for splits and cracks during timber drying and their control === The chief difficulty experienced in the drying of timber is the tendency of its outer layers to dry out more rapidly than the interior ones. If these layers are allowed to dry much below the fibre saturation point while the interior is still saturated, stresses (called drying stresses) are set up because the shrinkage of the outer layers is restricted by the wet interior. Rupture in the wood tissues occurs, and consequently splits and cracks occur if these stresses across the grain exceed the strength across the grain (fibre to fibre bonding). The successful control of drying defects in a drying process consists in maintaining a balance between the rate of evaporation of moisture from the surface and the rate of outward movement of moisture from the interior of the wood. The way in which drying can be controlled will now be explained. One of the most successful ways of wood drying or seasoning would be kiln drying, where the wood is placed into a kiln compartment in stacks and dried by steaming, and releasing the steam slowly.

The 1988 US presidential elections lent new urgency to the negotiations, which had recently stalled after six consecutive rounds of talks in Brazzaville. Angola and Cuba had gambled heavily on a victory for Michael Dukakis and the Democratic Party during the US elections, hoping that this would spell the end of US aid to UNITA and a harder line on South Africa. At the time of the Geneva Protocol, dos Santos had commented that "if the Democrats had won the elections, there would be a readjustment in US policy, particularly on Southern Africa". The election of Republican candidate George H. W. Bush had the effect of persuading the Angolan and Cuban delegations to be more flexible. Crocker reiterated on several occasions that a new US administration meant changes in personnel and basic policy review, and pressed them not to waste months of effort. Three days after the US election results were released, the parties reconvened in Geneva and within the week had agreed to a phased Cuban withdrawal over the course of twenty seven months. In exchange, South Africa pledged to begin bestowing independence on South West Africa by 1 November 1989. On 13 December 1988, South Africa, Angola, and Cuba signed the Brazzaville Protocol, which affirmed their commitment to these conditions and set up a Joint Military Monitoring Commission (JMMC) to supervise the disengagement in Angola. The JMMC was to include Soviet and US observers. All hostilities between the belligerents, including PLAN, were to formally cease by 1 April 1989.
nuclease Any of a class of enzymes capable of cleaving phosphodiester bonds connecting adjacent nucleotides in a nucleic acid molecule (the opposite of a ligase). Nucleases may nick one strand or cut both strands of a duplex molecule, and may cleave randomly or at specific recognition sequences. They are ubiquitous and imperative for normal cellular function, and are also widely employed in laboratory techniques.
== Medical uses == Flupirtine is used as an analgesic for acute pain, in moderate-to-severe cases. Its muscle relaxant properties make it popular for back pain and other orthopaedic uses, but it is also used for migraines, in oncology, postoperative care, and gynaecology. In 2013 due to issues with liver toxicity, the European Medicines Agency restricted its use to acute pain, for no more than two weeks, and only for people who cannot use other painkillers.
=== 13 June === The Ukrainian military claimed that its Third Special Operations Forces Regiment had destroyed a Russian R-416GM digital radio relay communication station, a truck-based system designed to improve the efficiency of radio relay units in the field, for the first time. Russia claimed to have shot down three drones over Yaroslavl Oblast and another over Vladimir Oblast. In Belgorod, the Governor claimed a drone damaged a car and injured two people. A Ukrainian court convicted a former executive of the Zaporizhzhia Nuclear Power Plant of collaborating with Russia and sentenced him to ten years' imprisonment. The G7 agreed to provide a $50 billion-loan derived from frozen Russian assets for military use by Ukraine during a summit in Italy that also saw the US and Ukraine sign a 10-year bilateral security deal. Canada pledged 2,000 decommissioned CRV7 “rocket motors”, armoured vehicles to be assembled in Kyiv, 29 Nanuk Remotely Controlled Weapon Stations and 130,000 rounds of ammunition to Ukraine.
== Detrital zircon data == Depending on the detrital zircon study, there should be different variables included for analysis. There are two main types of data, analyzed zircon data (quantifiable data and imagery/descriptive data), and sample (where they extract the zircon grains) data. Details are listed in Table 2.

Expectations diminished as a result of numerous failed replications, the retraction of several previously reported positive replications, and the identification of methodological flaws and experimental errors in the original study. By late 1989, most scientists considered cold fusion claims dead, and cold fusion subsequently gained a reputation as pathological science. In 1989 the United States Department of Energy (DOE) concluded that the reported results of excess heat did not present convincing evidence of a useful source of energy and decided against allocating funding specifically for cold fusion. A second DOE review in 2004, which looked at new research, reached similar conclusions and did not result in DOE funding of cold fusion. Presently, since articles about cold fusion are rarely published in peer-reviewed mainstream scientific journals, they do not attract the level of scrutiny expected for mainstream scientific publications. Some interest in cold fusion has continued through the decades—for example, a Google-funded failed replication attempt was published in a 2019 issue of Nature. A small community of researchers continues to investigate it, often under the alternative designations low-energy nuclear reactions (LENR) or condensed matter nuclear science (CMNS).
===== 1940s ===== In 1943, the 24 Hour Ration was devised as a direct replacement to the 48 Hour Mess tin ration. It contained only dried goods (no tins), to save weight and tinplate, which was the criticism of the earlier mess tin ration. It was first issued to troops on D-Day to provide interim food prior to supply lines being established which would permit 14 Man Composition Rations being brought ashore.
Positive social connections and interactions are further key elements of well-being. In addition to the intrinsic joy of engaging with others, social networks can offer material and emotional support during challenging times. They help people build trust, share values, promote the exchange of information, and provide access to new opportunities. Different types of social relationships may influence well-being in different ways, such as the contrasts between friends, family members, romantic partners, co-workers, and teammates. Researchers often focus specifically on the connection between friendship and well-being. Friendship is a voluntary social relationship between people characterized by mutual concern, trust, and support. Friends tend to spend time with each other, enjoy each other's company, and know personal facts about one another. Friendship can promote emotional support, a sense of belonging, self-esteem, practical guidance, and reduced stress. Different forms of friendship can influence well-being differently. A central aspect is the strength of the connection, distinguishing close friends from distant ones. Strength is determined by factors such as time spent together, trust, emotional intensity, and readiness to support each other in difficult times. The number of friends a person has is another relevant factor, and having many friends is usually beneficial. However, if a person already has numerous friends, making even more friends may not significantly impact their well-being.
The end of the Hundred Years War in 1453 meant that Britain, and thus Bristol, lost its access to Gascon wines and so imports of Spanish and Portuguese wines increased. Imports from Ireland included fish, hides and cloth (probably linen). Exports to Ireland included broadcloth, foodstuffs, clothing and metals. It has been suggested that the decline of Bristol's Iceland trade for 'stockfish' (freeze dried cod) was a hard blow to the local economy, encouraging Bristol merchants to turn west, launching unsuccessful voyages of exploration in the Atlantic by 1480 in search of the phantom island of Hy-Brazil. More recent research, however, has shown that the Iceland trade was never more than a minor part of Bristol's overseas trade and that the English fisheries off Iceland actually increased during the late 15th and 16th centuries. In 1487, when king Henry VII visited the city, the inhabitants complained about their economic decline. Such complaints, however, were not uncommon among corporations that wished to avoid paying taxes, or which hoped to secure concessions from the Crown. In reality, Bristol's customs accounts show that the port's trade was growing strongly during the last two decades of the fifteenth century. In great part this was due to the increase of trade with Spain.

Common methods include enzymatic recycling assays, liquid chromatography, and mass spectrometry. Many protocols separate reduced glutathione from its oxidized disulfide form before detection.
The ratio compares reduced glutathione with its oxidized dimer. It is used as an indicator of redox status, although the value depends strongly on sample handling and analytical method.
Glutathione can oxidize quickly after a sample is collected. Acidification, cooling, and chelators are often used to reduce artifactual changes before analysis.
Glutathione is built from three amino acids: glutamate, cysteine, and glycine. The linkage involves the gamma-carboxyl group of glutamate rather than the alpha-carboxyl group, which is unusual for peptides. This structure protects the bond from some common peptidases.
