== Myelin clearance == Myelin is a phospholipid membrane that wraps around axons to provide them with insulation. It is produced by Schwann cells in the PNS, and by oligodendrocytes in the CNS. Myelin clearance is the next step in Wallerian degeneration following axonal degeneration. The cleaning up of myelin debris is different for PNS and CNS. PNS is much faster and efficient at clearing myelin debris in comparison to CNS, and Schwann cells are the primary cause of this difference. Another key aspect is the change in permeability of the blood-tissue barrier in the two systems. In PNS, the permeability increases throughout the distal stump, but the barrier disruption in CNS is limited to just the site of injury.
transcript A product of transcription; that is, any RNA molecule which has been synthesized by RNA polymerase using a complementary DNA molecule as a template. When transcription is completed, transcripts separate from the DNA and become independent primary transcripts. Particularly in eukaryotes, multiple post-transcriptional modifications are usually necessary for raw transcripts to be converted into stable and persistent molecules, which are then described as mature, though not all transcribed RNAs undergo maturation. Many transcripts are accidental, spurious, incomplete, or defective; others are able to perform their functions immediately and without modification, such as certain non-coding RNAs.

== History == Sean Vanaman and Jake Rodkin were both at Telltale Games as co-writers on the 2012 game The Walking Dead, which was considered Telltale's first turn into a more narrative type of episodic adventure game. The game was a critical success, and while other projects came along, Vanaman started to become too comfortable with his role at Telltale, and felt that he would be more motivated if there was more discomfort or risk in his duties. After talking about the idea with Rodkin, the two left the company and co-founded Campo Santo on September 18, 2013, joined by Mark of the Ninja designer Nels Anderson and graphic artist Olly Moss. Environment artist Jane Ng and designer and composer Chris Remo, who had also worked with Vanaman and Rodkin on the Idle Thumbs podcast, soon joined the team as well. Soon after, they announced Panic would back their debut project. After a painting by Moss, Ng adapted the painting's aesthetic style into a 3D environment with the color and inspiration drawn from both New Deal advertisements and icons from the National Park Service as well as a camping trip in Yellowstone National Park in which the team visited a preserved fire lookout tower two miles (3.2 km) from the campsite. Development eventually led to the announcement of Firewatch in March 2014, with release originally slated for 2015. In August 2014, a game demo of Firewatch was released at PAX West, revealing the overall plotline and story of a fire lookout named Henry in the Shoshone National Forest in 1989.

=== Gene therapy prior to conception === Mitochondrial replacement therapy (MRT), where the nuclear DNA is transferred to another healthy egg cell leaving the defective mitochondrial DNA behind, is an IVF treatment procedure. Using a similar pronuclear transfer technique, researchers at Newcastle University led by Douglass Turnbull successfully transplanted healthy DNA in human eggs from women with mitochondrial disease into the eggs of women donors who were unaffected. In such cases, ethical questions have been raised regarding biological motherhood, since the child receives genes and gene regulatory molecules from two different women. Using genetic engineering in attempts to produce babies free of mitochondrial disease is controversial in some circles and raises important ethical issues. A male baby was born in Mexico in 2016 from a mother with Leigh syndrome using MRT. In September 2012 a public consultation was launched in the UK to explore the ethical issues involved. Human genetic engineering was used on a small scale to allow infertile women with genetic defects in their mitochondria to have children. In June 2013, the United Kingdom government agreed to develop legislation that would legalize the 'three-person IVF' procedure as a treatment to fix or eliminate mitochondrial diseases that are passed on from mother to child. The procedure could be offered from 29 October 2015 once regulations had been established.
=== Binding of substrates and co-substrates === Binding of phosphorylated substrates to distinct regions of the C-lobe has previously been detected by binding of a tungstate derivative (as a phosphate analog). Instead of phospho-primed substrate also the C-terminal regulatory domain of CK1δ is able to bind to this position for the purpose of autoregulatory function. Binding of ATP is mainly mediated via the glycine-rich P-loop (L-12, bridging strands β1 and β2), forming the top cover of the WTP binding site, and the so-called catalytic loop (L-67). Conformational changes affecting the activation loop (L-9D) are related to regulation of kinase activity. When the activation loop moves out of the catalytic site the catalytically relevant DFG motif (Asp-149, Phe-150, and Gly-151) shifts to an internal position. The aspartate residue chelates a Mg2+ ion allowing proper binding and orientation of ATP. Another residue, which is essentially involved in the regulation of kinase activity, but also in forming interactions with small molecule inhibitors, is Met-82, the so-called gatekeeper residue. Directly located within the ATP binding pocket this residue controls access of small molecules to certain binding pockets (selectivity pockets) located beyond the position of the gatekeeper.
== Regulation == There are many mechanisms that exist to regulate the expression of system Xc-, although it is not the sole determinant of extracellular glutamate or intracellular glutathione. An example is amino acid deprivation, which triggers up regulation of the transporter. A key regulator is extracellular glutamate; when it becomes excessive, it goes from an excitatory transmitter to an excitotoxin. The inhibition of uptake of extracellular cystine into cells leads to decreased levels of intracellular glutathione which leads to ferroptosis. This regulation may be done through Excitatory Amino Acid Transporters (EAATs), which decrease extracellular glutamate and increase intracellular glutamate in astrocytes. When looking at its structure, xCT seems to be the main determinant for the system's activity. Glutamate and cystine can be transported in both directions, but, generally, more cystine is imported and more glutamate is exported. Extracellular glutamate acts as a competitive inhibitor for cystine uptake via system Xc-.
